Glycosylation is a powerful enzymatic process that plays a pivotal role in all living organisms including prokaryotic cells. The modification controls numerous cellular processes including cell and protein stability and signaling (Ohtsubo & Marth, 2006). UDP NAM is an essential building block of bacterial peptidoglycan biosynthesis. While synthetic preparation of this molecule and derivatives thereof prove challenging, we outline here a robust and scalable step-by-step chemoenzymatic synthesis of UDP-NAM chemical probes. This process harnesses the promiscuity of bacterial cell wall recycling enzymes AmgK and MurU to generate a library of compounds that are of interest to the carbohydrate, microbiology and immunology communities studying nucleotide phosphates and their role in bacterial survival.
